A\nprevious study showed that the lightharvesting-\nreaction center core complex (LH1-\nRC) purified from this bacterium is highly\nstable at room temperature [Suzuki et al.,\n(2007) Biochim. Biophys. Acta 1767, 1057-1063].\nIn this work, we demonstrate that thermal\nstability of the Tch. tepidum LH1-RC is much\nhigher than that of its mesophilic counterparts\nand the enhanced thermal stability requires\nCa2+ as a cofactor. Removal of the Ca2+ from\nTch. tepidum LH1-RC resulted in a complex\nwith the same degree of thermal stability to\nthat of the LH1-RCs purified from mesophilic\nbacteria. The enhanced thermal stability can\nbe restored by addition of Ca2+ to the Ca2+-\ndepleted LH1-RC, and this process is fully\nreversible. Interchange of the thermal\nstability between the two forms is accompanied\nby a shift of the LH1 Qy transition between 915\nnm for the native and 880 nm for the Ca2+-\ndepleted LH1-RC. Differential scanning\ncalorimetry measurements reveal that\ndegradation temperature of the native LH1-RC\nis 15 °C higher and the enthalpy change is\nabout 28% larger than the Ca2+-depleted LH1-\nRC. Substitution of the Ca2+ with other metal\ncations caused a decrease in thermal stability of\nextent depending on the properties of the\ncations.
