We demonstrate in this study,\nusing a highly purified LH1-reaction center\ncomplex, that the LH1 Qy transition is strongly\ndependent on metal cations and Ca2+ is\ninvolved in the unusual red-shift. Removal of\nthe Ca2+ resulted in formation of a species with\nthe LH1 Qy absorption at 880 nm, and addition\nof the Ca2+ to the 880-nm species recovered the\nnative 915-nm form. Interchange between the\ntwo forms is fully reversible. Based on\nspectroscopic and isothermal titration\ncalorimetry analyses, the Ca2+-binding to the\nLH1 complex was estimated to occur in a\nstoichiometric ratio of Ca2+/αβ -subunit = 1:1\nand the binding constant was in 105 M-1 order of\nmagnitude, which is comparable with those for\nEF-hand Ca2+-binding proteins. Despite the\nhigh affinity, conformational changes in the\nLH1 complex upon Ca2+-binding were small\nand occurred slowly with a typical time\nconstant of about 6 minutes. Replacement of\nthe Ca2+ with other metal cations caused blueshifts\nof the Qy bands depending on the\nproperty of the cations, indicating that the\nbinding site is highly selective.
